Vitamin B6: Clinical Evidence & Substantiation Summary
What Is Vitamin B6?
Vitamin B6 (pyridoxine) is a water-soluble vitamin involved in over 100 enzymatic reactions, primarily in amino acid metabolism, neurotransmitter synthesis, and immune function. Pyridoxal 5'-phosphate (P5P) is the active coenzyme form.
Mechanism of action: P5P serves as a cofactor for aminotransferases, decarboxylases, and other enzymes in amino acid metabolism. It is essential for synthesis of serotonin, dopamine, GABA, and norepinephrine. B6 also modulates steroid hormone receptor expression and is required for hemoglobin synthesis.
Clinical Evidence Summary
Below are 5 key clinical studies on Vitamin B6. Nutra Comp analyses 56+ studies in its full clinical evidence report.
Vitamin B6 and depression: meta-analysis
Population: 14 studies
Key finding: Higher B6 intake was inversely associated with depression risk. Supplementation as adjunct therapy showed modest benefits (p=0.04).
B6 supplementation and PMS symptoms
Population: 9 RCTs
Key finding: B6 (50–100 mg/day) significantly reduced PMS symptoms including mood changes, irritability, and bloating (p<0.01).
Vitamin B6 and homocysteine
Population: 12 RCTs
Key finding: B6 supplementation reduced homocysteine levels, with greater effect when combined with folate and B12.
B6 and immune cell function
Population: 8 studies
Key finding: B6 deficiency impaired lymphocyte proliferation and IL-2 production. Supplementation restored immune parameters in deficient populations.
P5P vs pyridoxine: bioavailability comparison
Population: 36 healthy adults
Key finding: P5P and pyridoxine HCl showed similar overall bioavailability, but P5P provided higher plasma P5P levels in individuals with liver dysfunction.

Frequently Asked Questions
Is vitamin B6 FDA approved?
B6 is an essential vitamin sold as a dietary supplement. The Daily Value is 1.7 mg. It does not require FDA drug approval.
What are the most studied benefits?
Neurotransmitter support, PMS symptom relief, immune function, and homocysteine metabolism. Benefits are strongest in deficient populations.
What dosage is used?
10–100 mg/day depending on the indication. Long-term intake above 100 mg/day of pyridoxine carries risk of peripheral neuropathy. P5P may be preferred for those with liver concerns.
